In this study, thermodynamic aspects of Nifuratel (NFT) solubility improvement in the cyclodextrins solutions were thoroughly investigated. To this end, thermodynamic parameters of intrinsic solubility, dissolution, solubilization and complexation were experimentally delved and discussed in terms of driving forces of the outlined processes. The enthalpy driven and determined complexation was shown. A greater impact of hydrophobic effect to the complex formation of NFT with β-CD as compared to 2,6-Di-O-methyl-β-cyclodextrin (DIMEB) was estimated. Two-component and three-component solid dispersions (SD) of NFT with cyclodextrin (DIMEB) and polymer (PVP) were prepared and fully characterized. The dissolution/release and permeation rates of NFT, NFT/cyclodextrin and NFT/cyclodextrin/polymer SDs were measured using the side-by-side diffusion cell and artificial polydimethylsiloxane–polycarbonate membrane (PDS). The maximal increase in both the dissolution and permeation rates was achieved for the three-component NFT/DIMEB/PVP solid dispersion.
